Question 2014 M6GC w/Dinan Stage 1 Dyno Run

I will be putting my M6 on a Dyno this weekend for the first time ever. I have never done any Dyno runs, so I was curious as to best practice(s) for setting the car's suspension, shifting (DCT), acceleration sensitivity to get the most out of the car. Will a fully warmed up car run better than a car that has sat for an hour or so?

The weather is expected to be ~60-65F, Seattle standard non-rain humidity.
